PNC-27 is a synthetic chimeric peptide of 32 residues that joins the p53 residue 12 to 26 segment, which carries the HDM-2 binding domain, to a 17-residue membrane-penetrating leader sequence. It is produced through controlled solid-phase peptide synthesis and supplied as a lyophilized powder in a 10mg vial with no fillers added. The peptide is characterized in laboratory work on peptide interaction with HDM-2 associated with the plasma membrane. It is studied for membrane pore formation and for selectivity between transformed and non-transformed cultured cell lines.

PNC-27 belongs to the chimeric peptide class, in which a defined protein-binding motif is fused to a cell-penetrating or membrane-residency sequence. Its amino-terminal segment, PPLSQETFSDLWKLL, reproduces residues 12 to 26 of the p53 protein. Its carboxyl-terminal segment, KKWKMRRNQFWVKVQRG, supplies the membrane-penetrating leader.
The compound is prepared by solid-phase peptide synthesis and purified by reversed-phase HPLC. It carries no post-translational modification, no fluorophore, and no conjugated carrier in this format.
Research interest in PNC-27 centers on a physical chemistry question rather than a signaling one. The peptide is examined for how a p53-derived sequence adopts an HDM-2 binding conformation and what happens at a lipid bilayer when that binding partner is present in the membrane rather than the nucleus.
Soluble in water; aqueous solubility of ≥100 mg/mL has been reported for the free peptide
Storage
-20°C, protect from light
Storage and Handling
Store the lyophilized compound at -20°C in the sealed vial, protected from light and moisture.
After reconstitution, store at 2°C to 8°C and use promptly; for longer holds, aliquot and store at -20°C to avoid repeated freeze-thaw cycles.
Allow the vial to reach ambient temperature before opening so condensation does not enter the powder.
Maintain aseptic handling to preserve compound integrity.
Lyophilized Format
This compound ships in lyophilized (freeze-dried) form. Freeze-drying supports long-term storage stability and preserves compound integrity. No fillers are added.
PNC-27 is a 32-residue chimeric peptide that joins residues 12 to 26 of p53, the segment that binds HDM-2, to a membrane-residency sequence from penetratin. NMR work in aqueous and membrane-mimetic solvents found that it folds into amphipathic helix-coil-helix structures, with its hydrophobic residues gathered on one face [1].
The p53 residues of that structure superimpose on the same residues bound to HDM-2, which pointed researchers toward HDM-2 as the peptide’s binding partner at the cell surface [2]. HDM-2 was then detected in the membranes of several transformed cell lines but not in untransformed lines, and fluorescent labeling placed PNC-27 together with the membrane-bound protein [2].
Untransformed MCF-10-2A cells became susceptible to PNC-27 once they were made to express full-length HDM-2 with a membrane-localization signal [2]. A double-labeled form of the peptide stayed intact in MCF-7 cell membranes during lysis, while the untransformed MCF-10-2A line degraded it and remained viable [3].
The same dependence on membrane HDM-2 has been reported in the SKOV-3 and OVCAR-3 lines, with no colocalization or cytotoxicity in HUVEC controls [4]. Work in the p53-null K562 line points to a mechanism that does not rely on p53 activity inside the cell [5].
Later studies looked more closely at how the pores themselves assemble.
Pore Formation at the Plasma Membrane
Immuno-scanning electron microscopy found gold-labeled PNC-27 and HDM-2 in roughly 1:1 ratios within ring-shaped structures lining pores near the cell surface, and no pores formed in untransformed fibroblasts exposed to the peptide [6]. Pore assembly has been described as a temperature-independent 1:1 binding step followed by a temperature-dependent dimerization of those complexes [7].
In the MIA-PaCa-2 cell line, a monoclonal antibody against the p53 binding site of HDM-2 (residues 1 to 109) blocked PNC-27-induced necrosis, and immunogold labeling also placed the peptide on mitochondrial membranes [8].

A Certificate of Analysis (COA) is a document that verifies a compound’s identity, purity and quality through independent laboratory testing. Every compound from BioLongevity Labs is third-party tested, and its COA is published here, so researchers can review the testing record before a compound enters a protocol.
Each COA reports results from third-party laboratory analysis, including:
Ultra-high-performance liquid chromatography with mass spectrometry (UHPLC-MS) for purity, measured against a 98% or 99% specification depending on the compound
Mass identification for molecular confirmation and content quantitation
Endotoxin testing by Limulus amebocyte lysate (LAL) or recombinant Factor C (rFC) assay, depending on the laboratory
Visual and physical characterization of the finished material
How to Verify a COA Independently
You can check every certificate against the issuing laboratory’s own records, not just the copy hosted here. Verification does not depend on BioLongevity Labs.
AV Analytics certificates carry a search code. Scan the QR code on the certificate to open the laboratory’s verification page, or enter the search code at avanalytical.com/coa-search to pull the official record.
BioRegen reports of analysis carry a Report ID and a Validation Key. Scan the QR code on the certificate to open the official record, or quote both identifiers when you contact the laboratory at the address printed on the report.
SafeCert Labs issued a number of earlier certificates. Each one carries a COA number and the signature of the reporting chemist. Quote that number when you ask the laboratory to confirm it.
Older certificates from other laboratories list their own verification details on the document.
When a batch is sent to more than one laboratory, each certificate resolves at its own issuing laboratory. A researcher can then check the results against two unrelated sources.
These COAs come from independent certified laboratories, not in-house testing. That gives researchers a verifiable record of molecular integrity for each compound. All compounds are supplied for research use only.
